Comparative Analysis of Long-term Cultivation of Primary Epithelial Cells:
In our comparative analysis primary human keratinocytes were cultured in CnT-NX-EX medium, its predecessors CnT-07 and CnT-PR and three other commercially available media. Only keratinocytes grown in CnT-NX-EX achieved over 70 population doublings (at passage 19 since isolation) and their lifespan was extended beyond 100 days in culture. CnT-NX-EX outcompeted all other tested media with regards to proliferation rate, cell yield and longevity.
Prolonged Cultivation of Primary Human Juvenile and Adult Keratinocytes:
In our long-term culture assay CnT-NX-EX supported beyond 90 population doublings (at passage 21) in juvenile keratinocytes and beyond 60 population doublings (at passage 18) in adult keratinocytes. CnT-NX-EX significantly expanded lifespan of juvenile keratinocytes beyond 118 days and that of adult keratinocytes beyond 103 days in culture.
Colony Forming Efficiency of Adult Keratinocytes at Higher Passages:
Adult keratinocytes cultured in CnT-NX-EX, its predecessors CnT-07 and CnT-PR and three other commercially available media. Keratinocytes cultured in CnT-NX-EX showed excellent colony forming efficiency and outcompeted other tested media by 1.5-fold. Colonies were not observed for other tested media beyond the fourth passage. Whereas, with CnT-NX-EX colonies were observed even at higher passages 14 and 19.
In vitro 3D Epidermal Modeling:
Adult keratinocytes cultured in CnT-NX-EX, its predecessors CnT-07 and CnT-PR and three other commercially available media. Epidermal models were created by 3D air-liquid interface culture using CnT-PR-3D medium. Left panel: 3D epidermal models created with adult keratinocytes cultured in CnT-NX-EX showed better performance as compared to other tested media. Right panel: 3D epidermal models with adult keratinocytes expanded in CnT-NX-EX at passage 14 showed excellent cell morphology. No other tested media were able to create a well differentiated 3D model at higher passages.
