CnT-AG2 (VitroAge) is a chemically defined medium specifically designed to age epidermal keratinocytes over several passages without the need for artificial pro-aging treatments such as peroxide or other oxidizing agents. Unlike traditional culture media—which have evolved to promote strong proliferation and contain protective and stimulatory factors that exert powerful anti-aging effects—CnT-AG2 lacks these protective elements. As a result, keratinocytes age naturally in this medium over a period of several weeks.
The VitroAge epidermal model maintains normal cell morphology but develops well-documented in vivo signs of aging, including reduced lifespan and disrupted metabolism. Research shows that aged epidermis is characterized by a reduced number and function of transient-amplifying progenitor cells (Charruyer et al., JID 2009), which cycle more slowly and fewer times in aged tissue. A similar reduction in cell cycle kinetics is observed in VitroAge cultures during four weeks of aging.
Keratinocytes aged in CnT-AG2 exhibit a range of additional age-related changes, making this medium ideal for studies on skin aging and related processes. CnT-AG2 is fully defined, does not contain antibiotics/antimycotics, and provides a controlled environment for natural aging without external chemical stimuli.
