The CnT‑FT‑CELLS Full‑Thickness Primary Cell Value Pack contains two primary cell types required for establishing full‑thickness 3D skin models: (i) normal human epidermal keratinocyte progenitor cells (HPEKp) isolated from juvenile donor skin, and (ii) normal human dermal fibroblasts (HDFp) isolated from juvenile dermal tissue. These cells have been successfully used with CnT‑NX‑E, CnT‑PR‑F, and CnT‑PR‑FTAL5 media to generate fully stratified full‑thickness skin models.
Because both cell types are of human origin, they must be handled as hazardous. Although they have been tested negative for HIV‑1, Hepatitis B, and Hepatitis C, they should be treated as potentially infectious. Appropriate biocontainment procedures, protective equipment, and laboratory safety measures must be employed to avoid accidental exposure.
For successful model establishment, the keratinocytes and fibroblasts should first be expanded for one to two passages in their respective proliferation media. HPEK cells should be expanded in CnT‑NX-E medium, while HDF cells should be expanded in CnT‑PR‑F medium prior to seeding into full‑thickness culture. Thawing, seeding, and passaging procedures play an important role in achieving optimal cell growth, and users are advised to follow the recommended protocols available on the CELLnTEC Protocols Page.
