Description

CnT-PR-AD is a fully defined, chemically formulated medium designed for the differentiation of large airway epithelial cells in 2D or 3D culture. It is completely free of animal or human-derived components and does not contain phenol red or antibiotics/antimycotics. This medium is the evolution and refinement of our previous airway differentiation formulation, CnT-23.
CnT-PR-AD contains a specifically tailored mix of growth factors, where proliferative components that retard differentiation are omitted or reduced, and pro-differentiation components are added. It uses an optimized basal medium enriched with trace elements, protective antioxidants, and vitamins to create an environment conducive to full differentiation. PCT growth factors and other proliferative factors are intentionally excluded to promote differentiation.
The formulation is low-calcium, as calcium plays a key role in the differentiation of confluent epithelial cells. It is recommended to add calcium once the cells are fully confluent—please refer to our differentiation protocol for detailed guidance.
CnT-PR-AD is used for differentiating airway epithelial cells that have been expanded in CnT-PR-A or CnT-17 media.

It is strongly recommended to use airway epithelial cells that are growing in CELLnTEC aiway epithelial proliferation media such as CnT-17 or CnT-PR-A, to avoid a switch in base medium composition during model formation.
