The PR2D‑CC 2D Epithelial/Stromal Co‑Culture Starter Kit provides all components required to establish a balanced co‑culture of primary keratinocytes and fibroblasts in a fully defined environment. The kit includes primary human keratinocyte and fibroblast cells, CnT‑PR and CnT‑PR‑F media for monolayer expansion, and CnT‑PR‑CC co‑culture medium. It is designed to enable straightforward and reliable establishment of 2D co‑culture models in the laboratory.
Because the cells included in the kit are of human origin, they must be handled as hazardous. Although they have been tested negative for HIV‑1, Hepatitis B, and Hepatitis C, they must be treated as potentially infectious, and appropriate biocontainment procedures, protective equipment, and laboratory precautions should be applied to prevent accidental exposure.
The included primary keratinocytes (HPEKp) and fibroblasts (HDFp) are first expanded in their respective proliferation media following the recommended general protocol. After expansion, both cell types are seeded at an equivalent ratio into standard 2D culture using CnT‑PR‑CC co‑culture medium, where they proliferate together as a balanced co‑culture. CnT‑PR‑CC is a fully defined, serum‑ and BPE‑free formulation that omits fibroblast inhibitory elements, enabling fibroblasts to grow alongside keratinocytes under defined conditions.
At confluency, the co‑culture displays the characteristic small, tight cobblestone morphology of keratinocyte progenitor colonies surrounded by fibroblasts, reflecting a balanced epithelial–stromal interaction in vitro.
