The PR3D‑HPEK‑50 3D Keratinocyte Starter Kit provides everything required to establish three‑dimensional epidermal models with improved differentiation and barrier function in the laboratory. The kit includes primary human keratinocytes (HPEKp), CnT‑NX‑E epithelial culture medium for monolayer expansion, CnT‑PR‑3D airlift medium for 3D differentiation, and forty‑eight 0.4 μm pore‑size cell culture inserts for air‑lift culture. Together, these components enable reliable and reproducible generation of 3D epidermal tissues.
Because the keratinocytes included in the kit are of human origin, they must be handled as hazardous. Although the cells have been tested negative for HIV‑1, Hepatitis B, and Hepatitis C, they should be treated as potentially infectious. Appropriate biocontainment procedures, protective equipment, and laboratory safety precautions must be followed to prevent accidental exposure.
Using the recommended protocol, the primary keratinocytes are first expanded in CnT‑NX‑E medium and then seeded onto culture inserts before transitioning to CnT‑PR‑3D airlift medium for stratification and barrier formation. When generated according to this protocol, the resulting 3D epidermal models routinely achieve up to 50% better barrier function compared to models produced using traditional media.
