Description

CnT-NX-EX is a next-generation medium designed for the efficient isolation and prolonged expansion of primary epithelial cells from various epithelial tissue types, including keratinocytes from skin, corneal epithelial cells, and oral epithelial cells, and other epithelial subtypes. CnT-NX-EX significantly enhances derivation and expansion efficiency. CnT-NX-EX medium is ROCK inhibitor-free, chemically defined, and contains no components of animal or human origin. No feeder cells or plate coating are required. The design ensures highest reproducibility and excellent inter-experimental and lot-to-lot consistency. CnT-NX-EX supports high cell proliferation rates while mitigating premature cellular senescence. Cells grown in CnT-NX-EX exhibit excellent morphology and maintain functionality and ability to differentiate into all epithelial layers up to highest passage numbers.

CnT-NX-EX

Primary human adult keratinocytes cultured at higher passages 6 and 18 in the new CnT-NX-EX shows superior growth and impeccable cell morphology.

CnT-NX-EX is a next-generation medium designed for the efficient isolation and prolonged expansion of primary epithelial cells from various epithelial tissue types, including keratinocytes from skin, corneal epithelial cells, and oral epithelial cells, and other epithelial subtypes.

Compared to other commercially available formats, CnT-NX-EX significantly enhances derivation and expansion efficiency. The medium achieves up to 90 population doublings in human neonatal keratinocytes and 60 doublings in adult keratinocytes, with the potential to achieve up to a trillion-fold increase in cell numbers over the duration of the culture.

More efficient cell expansion allows you to perform repeats and technical replicates from the same sample and avoid sourcing additional cells or patient material. The increase in population doublings and cellular longevity means that you can significantly increase experimental or assay time windows to up to 100 days or more.

CnT-NX-EX supports high cell proliferation rates while mitigating premature cellular senescence. Cells grown in CnT-NX-EX exhibit excellent morphology and maintain functionality and ability to differentiate into all epithelial layers up to highest passage numbers.

CnT-NX-EX medium is ROCK inhibitor-free, chemically defined, and contains no components of animal or human origin. No feeder cells or plate coating are required. The design ensures highest reproducibility and excellent inter-experimental and lot-to-lot consistency. This medium is fully supplemented and comes in a single bottle, ready-to-go.

The medium is ideally suited to support in vitro studies on skin and inflammatory diseases, irritation assays, and cosmetic or drug evaluations, and other 3D-modelling applications. It enables a seamless transition from lab to clinical trial and ATMP-development.

Long-term Cultivation of Human Neonatal and Adult Keratinocytes: CnT-NX-EX supports up to 90 population doublings (PDs) in neonatal keratinocytes which is significantly higher than 40 PDs obtained with predecessor CnT-PR. Likewise, CnT-NX-EX supports up to 70 PDs in adult keratinocytes as compared to 25 PDs obtained with CnT-PR.

 

Comparative Analysis of Long-term Cultivation of Adult Human Keratinocytes: Primary cells were cultured in CnT-NX-EX medium, and its predecessors CnT-07 and CnT-PR and three other commercially available medium. Only keratinocytes grown in CnT-NX-EX showed enhanced proliferation rates, achieved a higher number of population doublings of up to 70 PDs, and extended the lifespan of keratinocytes for up to 110 days (and beyond) in culture. CnT-NX-EX outcompeted all other tested media.

CnT-NX-EX_3D

Primary epithelial cells growing in CnT-NX-EX display a highly proliferative phenotype. For differentiation experiments, it is recommended to switch to the CnT-PR-D or CnT-PR-3D differentiation media. CnT-NX-EX supports 3D model creation at higher passages which is not possible by conventional media. In vitro 3D epidermal models with adult keratinocytes expanded in CnT-NX-EX medium at higher passages shows excellent cell morphology as seen in the figure.
