The VitroAge medium (CnT-AG2) is specifically designed to age epidermal keratinocytes over several passages, without the need for artificial pro-aging treatments such as perioxide.

Traditional culture media have evolved over 50 years to provide strong proliferation for extended periods. They contain many protective and stimulatory factors and nutrients, that exert a powerful anti-aging effect.

CnT-AG2 lacks these protective elements. Keratinocytes age naturally in this medium over a period of several weeks, without the need for acute doses of oxidizers or other synthetic pro-aging chemicals.

The Vitroage epidermal model maintains normal cell morphology, but generates several well documented in vivo signs of ageing including reduced lifespan and disrupted metabolism. For an overview of key ageing-related literature, please click here.

It is now well documented that the aged epidermis is characterized by a reduced number and function of transient-amplifying progenitor cells (Charruyer et al, JID 2009). In particular, these progenitors were found to cycle more slowly, and fewer times in aged tissue. A similar reduction in cell cycle kinetics is also observed in VitroAge during 4 weeks of culture (see Fig at left).

VitroAge Keratinocyte Aging Medium

CnT-AG2 is a fully defined, low calcium (0.07 mM) formulation designed to age keratinocytes over several weeks without external chemical stimuli or oxidisers. It does not contain antibiotics / antimycotics. The basal medium contains amino acids, minerals, vitamins and organic compounds, but is protein free. 

Developed for human keratinocytes. May be used for other species as well (not yet tested).

Product use
Developed specifically to age keratinocytes over several weeks in culture. Keratinocytes should be growing in CnT-Prime medium prior to switching to CnT-AG2 medium.  For proper cell recovery and efficient cell pelleting during passaging, additional protein (such as BSA) must be added to the medium during centrifugation. This medium is designed for use in a 5% CO2 atmosphere. For suggested cell culture protocols, please see our resources section.
For routine cell cultivation, CELLnTEC recommends to work without antibiotics / antimycotics.
Storage / Shelf life
Store frozen below -15 °C. For best before date, see label. To prepare medium for use, thaw in a water bath set to room temperature. Do not use higher temperatures. Swirl frequently, approximately every 20 min, to ensure good mixing of the ingredients and temperature equilibration. Stop at melting of the last bit of ice to prevent warming at the end of the thawing process. Once thawed, medium has a remaining shelf-life of 6 weeks when stored at 4 °C in the dark. Certain culture media components are very sensitive to light. Minimize light exposure at all times.
Quality control
Media functionality and cell morphology tested by cell cultivation over 2 passages in supplemented medium. Osmolality, pH and concentration of various ions are tested. Free of bacteria, fungi and mycoplasma contamination.
